【Measurement of Ribose】
1. Ribose is dehydrated and cyclized in concentrated acid to form furfural, which reacts with orcinol (3,5-dihydroxytoluene) to show blue-green color and has a maximum absorption value at 670nm.
2. Reagents
Orcinol reagent: Combine orcinol (also known as lichenol, if it is a commercially available product that is oxidized to red, it needs to be recrystallized with hot benzene before use), ferric ammonium sulfate (ferric chloride can also be used instead) 13.5g, make 500mL with distilled water and store at low temperature. Before use, take 2.5mL of the stock solution, add 41.5mL of 12mol/L hydrochloric acid, and add distilled water to 50mL, which is the orcinol application solution.
3. Measurement
Take 1 mL of sample solution, add 3 mL of orcinol application solution, heat in a boiling water bath for 20 minutes, immediately cool with cold water, zero with reagent blank, measure the A670nm value, compare with the standard sample, and calculate the ribose content in the sample.
4. Description
Proteins and mucopolysaccharides may interfere with the determination.
